The insulation on your brain’s wiring wears out with age
Your brain runs on wiring. Billions of nerve fibers carry fast signals. Each one wears a coat called myelin — the insulation that keeps a signal sharp. Good myelin, clean message. Frayed myelin, slow or crossed message.
Let me explain what a team in Edinburgh just found. They studied hundreds of older Scots, tracked since birth in 1936, into their seventies and eighties. The people with the worst memory decline shared a strange thing. Not too little myelin. Too much of the wrong kind.
The cells that make myelin go faulty with age. They pile on thick, junk insulation. And they let the big, fast fibers wither. Picture old house wiring — lumpy tape in some spots, bare wire in others. The current still runs. It just runs badly.
  The team traced it to a drop in one protective protein inside those cells. Put that protein back in mice, and the faulty pattern started to reverse. Drugs that hit the same target already exist — for multiple sclerosis.
This was mice and human tissue, not a treatment yet. I could be wrong about how fast it moves. But I believe repair — not just prevention — is where this field is heading. And that is new.
Take a hard look at this one. It hints that an aging brain may be fixable, not only protectable.

This one is sobering. Researchers looked at 142 former professional soccer players and compared them to men their age. Nearly a third scored in the range for clinical depression. In the comparison group it was under one in ten. Their scans also showed less volume in areas for memory and attention. Years of heading the ball may be part of it.

Fish oil reached the brain and still fell short
Millions take fish oil for memory. So a two-year trial put it to a hard test — 365 older adults, many carrying the main Alzheimer’s risk gene. The high dose of omega-3 clearly reached the brain; levels in the spinal fluid rose 17%. But memory scores did not budge. And the memory center kept shrinking at the same pace.

A nerve from the gut woke up old memories
Here is an odd one from the gut. In aging mice, the line running from the belly up to the brain — the vagus nerve — had gone quiet. Switch it back on, and forgetful old mice ran mazes like young ones. Shifting gut bacteria seemed to jam the signal. It is mice for now, not people. But the gut-brain link keeps getting harder to ignore.

Here is where new treatments stand. The hunt has widened. Most drugs in testing now aim at something other than the sticky amyloid plaques — roughly four in five. Three from the newest round stood out.
A drug that shields the connections between brain cells
Brain cells talk across tiny junctions called synapses. Lose those, and memory goes. Zervimesine, from Cognition Therapeutics, is a pill meant to guard them — it blocks a docking site that lets toxic bits jam the junction. In a mid-stage trial it slowed symptoms in a related dementia. It is now heading toward a large, final-stage test in Alzheimer’s.

A drug that tells brain cells to make less of the tangle protein
Inside a failing neuron, a protein called tau clumps into tangles. Diranersen, from Biogen, is a genetic medicine that turns tau production down at the source. Its mid-stage trial in 416 people was mixed — it missed its main goal, but it lowered tau and slowed decline on most measures. It is moving to a final-stage test.

A cannabis-based medicine that calms end-stage agitation
Late dementia often brings severe agitation — hard on patients and families both. A trial called LiBBY tested a measured dose of two cannabis compounds, twice a day, in 120 people in hospice care. By twelve weeks, 87% had improved, against 24% on a dummy pill. This one is about comfort and dignity, not a cure.
